1. 2x LB에 접종해서 OD 0.6-0.8정도 까지 키우고 얼음에 20분 방치

2. sorvall tube로 옮겨 담고, 3600rpm, 4'C 5min 원심분리

3. cell down된 pellet을 glycerol을 이용하여 concentration(잔여배지를 없에기 위한 농축과정)

200ml ; glycerol 20 resuspend - 5min 원심분리 - glycerol 10 - 5min resuspend - glycerol 5 - 5min - glycerol 2

100ml ; 10-5-3-1

4. tube에 50-100씩 덜어서 액체질소 보관
