Slica gel에 DNA가 binding 하는 이유

Binding to silica surfaces in the presence of certain salts and under certain pH conditions.

negative를 띠는 slica gel과 DNA가 염을 사이에 두고 결합을 하게 되어 membrane에 binding하게 된다 그 후 증류수로 washing 하면 염이 없어서 다시 빠져나간다.

버퍼기능

1.S1 buffer : cell 현탁 solution, RNase 첨가, glucose가 삼투유지

2.S2 buffer : cell lysis solution, SDS가 세포벽에 구멍을 내고, NaOH가 .이중가닥 DNA를 단일가닥으로 분리

3.G3 buffer : 중화 solution, 산성인 아세틱 acid가 용액을 중화

4.PW buffer : Washing buffer, 80%이상의 에탄올이 genetic DNA를 침전
