단백질발현실험

5. cell harvest : large scale - 500ml bottle 두개에 옮겨 담아서 원심분리 후 냉동보관

5. cell harvest : small scale - 500 마이크로L 만 따로 떠서 원심분리

6. cell Lysis : small scale ; lysis 용액인 bugbuster 100 마이크로L 에 현탁 후 30분 상온보관, 원심분리 10분(!)

7. protein gel loading : SDS-PAGE ;

-soluble과 insoluble+buffer 100마이크로L 준비

-protein 20 마이크로L + 5*sample buffer 5 마이크로L

-HEAT ; denature 5min at 95`c

-1초 spin down

-marker랑 같이 로딩 ;

200V, 50min

-instant blue로 염색

-로딩결과 원하는 단백질로 추정되는 soluble protein이 보임

cf) 원심분리(!) 하는 이유 : 단백질이 제대로 발현됬으면 soluble이기 때문에, 그리고 변성을 막기위해 4`C 쿨링 centrifuge 로
